Viruses are supramolecular assemblies composed of proteins, nucleic acids, and sometimes lipids. Many highly pathogenic human RNA viruses exist, including influenza, Ebola, measles, and rabies viruses. Unlike many other viruses with rigid capsids, enveloped RNA viruses possess flexible and fragile architectures. As a result, elucidating their detailed structures has been challenging, and their assembly processes remain poorly understood. Recent advances in structural biology have enabled the determination of the structures of challenging biomolecular targets, including viruses. By integrating interdisciplinary approaches, combining virology and structural biology, we aim to uncover the assembly mechanisms of RNA viruses.
ウイルスの構造学
ウイルスは、タンパク質、核酸、そして場合によっては脂質から構成される超分子複合体です。インフルエンザウイルス、エボラウイルス、麻疹ウイルス、狂犬病ウイルスなど、ヒトに高い病原性を示すRNAウイルスが数多く存在します。多くのウイルスが硬いウイルス殻を持つのに対し、エンベロープを有するRNAウイルスは、柔軟で壊れやすい構造をしています。そのため、これらの詳細な構造を明らかにすることは難しく、またウイルス粒子がどのように形成されるのかについても、十分には解明されていません。 しかしながら、近年の生命科学技術の進展により、ウイルスを含む解析が困難とされてきた生体分子の構造が次々と明らかになってきています。私たちは、ウイルス学や構造生物学や融合させた学際的な手法を取り入れながら、RNAウイルスの粒子がどのように組み立てられるのか、その仕組みを包括的に解明することを目指しています。
