Viruses are supermolecular complexes consisting of proteins, nucleic acids, and sometimes lipids. Many highly pathogenic human RNA viruses exist, such as influenza, Ebola, measles, and rabies viruses. Unlike many other viruses with rigid viral shells, RNA viruses with envelopes possess flexible and fragile structures. Consequently, elucidating the detailed structure of these RNA viruses has been challenging, and their assembly processes remain largely unknown. Recent technological advancements in structural biology have enabled the clarification of structures for challenging biomolecular targets, including viruses. By combining interdisciplinary approaches such as virology and structural biology, we aim to unravel the assembly mechanisms of RNA viruses.
ウイルスは, 蛋白質・核酸・脂質が複雑に相互作用して形成される超分子複合体です. そのなかでもRNAゲノムをもつウイルスには, インフルエンザウイルス, エボラウイルス, 麻疹ウイルス, 狂犬病ウイルスなど, ヒトに対して高い病原性を示すウイルスが多数含まれています. 多くのウイルスが堅牢なウイルス殻を持つのに対し, RNAウイルスは脂質膜に包まれた柔らかく壊れやすい構造を持っています. そのため, RNAウイルス粒子の詳細な構造を捉えることが難しく, ウイルス粒子がどのように形成されるのかはよく判っていません. しかし，近年の構造解析技術の発展により，ウイルスを含めて今まで解析が難しかった生体分子の構造が次々に明らかになっています. 私たちはウイルス学や構造生物学などの学際的な研究手法を組み合わせてRNAウイルス粒子の形成機構を統合的に明らかにすることを目指しています.
